2 vs 3
I bought 2 mice and for the most part they seem to get along fine, possibly a little squabbling, has any one noticed a difference between 2 and 3 mice? there are still mice available from where i got them from their litter, if i got another one would they get along still or would i need to go about re-introducing them?

Really, it depends on when you got your pets. I'd say its unlikely for them to fight severely with litter mates unless they'd been separated for a long period of time.
Three mice finishes the colony, so its recommended, however two should be fine.
To be on the safe side, I'd introduce them slowly anyhow.

Females tend to get along whether or not they're from the same litter, as long as they're introduced correctly. Here's some information about how to do that: http://petmousefanciers.forumotion.com/t39-introducing-mice
Three females is often recommended because the relationship between two females can get too intense, and a third can serve as a sort of peacekeeper. Additionally, three means that if one passes away, no one will be left alone.

It "finishes" the colony as there are three main positions in a mouse colony. This doesn't mean you can't have more than 3; you absolutely can! Its just that those positions are needed for a colony. Anyone remember what these positions are called?

I don't remember what words we "officially" use for them, but they're basically like a dominant mouse, a submissive mouse, and a peace keeper.

Re: 2 vs 3
Three is absolutely better than two. Three mice make a complete colony, and colonies of only two girls can lead to over-dominance and bickering. However, if you get a third mouse, she will have to go through a three-week quarantine and you'll have to follow introduction procedures. Two mice can work, but if you notice them starting to argue, getting a third girl can really help.
